Firefly-luciferase
Description
Luciferase is a kind of enzyme that can catalyze the oxidation of luciferin to oxyluciferin and then emit bioluminescence. There is no endogenous luciferase in mammalian cell. Firefly-luciferase is a typical enzyme protein consisting of 550 amino acids, and functional luciferase is produced as soon as transcription is complete.
At present, luciferase assay is the ideal reporter gene detection method. The 3’UTR region of the target gene was constructed behind the luciferase gene in the vector, and then it was transfected into cells. The expression level of the target gene can be quantified by comparing the expression level of the reporter gene (monitoring the change of luciferase activity).
